Treatment of fresh root at low temperature was found to considerably increase the yield of methanolic extract of root of Panax ginseng C.A. MEYER harvested in July-November. The yield of methanolic extract of the root harvested in August, which had been treated at 2°± 1° for 30 days, was about 2.6 times that from untreated root. The quantitative determination of the constituents, i.e., dammarane-saponins (by GLC), sucrose (by HPLC and ^<13>C-NMR), amino acids (by amino acid analyzer), and ether-soluble substances showed that the increase of the yield of the extract is mainly due to the stimulation of sucrose production by this low temperature treatment. Thus in the extracts of the root treated with low temperature, the active principles such as saponins are evidently diluted with larger quantity of sucrose contained. The time course changes of sucrose and saponin contents in the extract during the low temperature-treatment were also investigated.
