Ginseng Radix is an indispensable herb medicine in traditional Chinese medicine and has long been used as a tonic of characteristic sort for treatment of miscellaneous diseases. It is clinically used popularly in the domain of Kampo medicine in Japan today. Although twenty-five saponins have been isolated and identified in Ginseng Radix, the quality evaluation of Ginseng Radix has been practically done according to the analysis of its main saponins such as ginsenoside-Rb_1 and -Rg_1. An assay system by HPLC was established, by which twenty-one saponins in a Ginseng Radix extract could be determined simultaneously. By this system each saponin content in the radixes of different growing years, different tissues of Ginseng Radix and two Ginseng Radix of different preparations (Red Ginseng and White Ginseng) were determined. The results showed that the total saponin contents in the cultivated Panax ginseng, increased until the third year, once decreased in the forth year and increased again from the fifth year, that Ro alone was contained in a higher level in parts of xylem and rhizome, and that heating with steam to process Panax ginseng into Red Ginseng, increased the number of saponins detected from eleven to sixteen. This assay manner is anticipated to be useful for the quality control of Ginseng Radix. Quality evaluation of Red Ginseng and White Ginseng by pharmacological means showed that the former had a stronger immunoinvigorative effect and blood flow promotive effect than the later. Red ginseng contain some water-soluble constituents produced in its repairing process, which may be considered to stimulate absorption of saponins into body besides the difference of their saponin levels. It is reliable that these findings obtained by chemical and pharmacological evaluations on Ginseng Radix supply a valuable indication for its practical application in the future.
