Fluorescence in situ hybridization using a high sensitive chemifluorescent substrate newly synthesized for horseradish peroxidase

For signal-generating systems mediated by enzymes in fluorescence in situ hybridization (FISH), eleven fluorogenic substrates of phenol derivatives for horseradish peroxidase (HRP) were synthesized. Among them, N-(4-hydroxy-2-methylphenyl)benzamide (HMPB) appeared to be the most useful and efficient substrate, and generated a bluish green fluorescent precipitate at the site of probe hybridized on chromosomes and nuclei with high stability and slow bleaching. Here, a new signal-generating system of the HRP/HMPB method for FISH was developed. This system could be used for gene assignment and dual-color FISH combined with the other method of ALP/HNPP-Fast Red TR procedure for the simultaneous visualization of different DNA sequences.
