Detection of the bovine Y specific DNA by in situ hybridization

Applying fluorescence in situ hybridization (FISH) with HNPP-azo dye and PCR products of bovine Y specific primer (BC 1.2) as a probe we detected the bovine Y specific DNA targets in metaphase spreads and in interphase nuclei derived from bovine lymphocytes. Hybrid signals, usually examined under a fluorescent microscope, were visualized in red color under a bright field light microscope. The present technique, therefore, would provide an additional and efficient system for embryo sexing.
