Molecular characterization of a t(11;22)(q24;q12) translocation junction in two cases of Ewing's sarcoma

The genomic and cDNA breakpoint junctions of a t(11;22)(q24;q12) translocation, which joins the EWS gene located on chromosome 22 with the FLI1 gene located on chromosome 11, were characterized with polymerase chain reaction (PCR)-based nucleotide sequencing in two cases of Ewing's sarcoma. RNA-PCR gave cDNA products from EWS-FLI1 chimeric gene, in which EWS exon 10 was fused with FLI1 exon 8 in the first case and FLI1 exon 5 in the second case. Genomic breakpoints in both the EWS-FLI1 and FLI1-EWS chimeric genes were found to occur in EWS intron 10 in both cases, joining with FLI1 intron 7 in the first case and with FLI1 intron 4 in the second case, respectively. Deletion and/or duplication of both genes, upto about 200bp, were found at the junctions on chromosomes 11 and 22 in these cases. The present results suggest that t(11;22) translocation in Ewing's sarcoma constitutes another example of asymmetric junctions at the molecular level, in addition to those reported previously in hematologic malignancies and solid tumors.
