Effects of Muscarinic Cholinergic Antagonists or p-Chlorophenylalanine on the Morphine-Induced Increase in Striatal Homovanillic Acid Concentration of Rats

Muscarinic cholinergic antagonists such as atropine and scopolamine significantly inhibited both the increase in locomotor activity and the rise in striatal HVA content induced by the administration of lower doses of morphine. p-CPA, an inhibitor of tryptophan 5-hydroxylase, on the other hand, significantly prevented both the decrease in locomotor activity and elevation of striatal HVA level caused by higher doses of morphine. Results suggest that the increase in cholinergic activity followed by elevation of dopaminergic activity in the striatum may be involved in an increase in locomotor activity by lower doses of morphine. In contrast, the decrease in locomotor activity by higher doses of morphine may be mediated by the rise in serotonergic activity and subsequent inhibitory effects on postsynaptic sites of dopaminergic neurons.
