Effects of 5, 7-Dihydroxytryptamine on Brain Monoamine Contents and on Morphine-Induced Hypothermia in Rats

The administration of p-chlorophenylalanine (p-CPA) (320mg/kg i. p. ) caused a prominent reduction in 5-hydroxytryptamine (5-HT), no significant change in norepinephrine (NE), and a slight but significant decrease in dopamine (DA) levels in the rat brain. The intestinal 5-HT concentrations were also significantly reduced following p-CPA injection. 5, 7-Dihydroxytryptamine (5, 7-DHT), injected intraventricularly at a dose of 100μg into rats pretreated with desmethylimipramine (DMI), produced a marked reduction in the brain 5-HT contents without any significant changes both in the brain catecholamine concentrations and in the intestinal 5-HT levels. The magnitude of the reduction in hypothalamic 5-HT contents by p-CPA administration was significantly greater than that following DMI-5, 7-DHT treatment. The administration of morphine (20mg/kg s. c. ) to rats produced prominent hypothermia, which was completely prevented by pretreatment of animals with p-CPA and partially but significantly inhibited by DMI-5, 7-DHT pretreatment. These results provide additional evidence of the involvement of 5-HT in morphine-induced hypothermia.
