Tissue Localization of T-and B-Lymphocytes in Humans

Lymphocyte subpopulations were identified on human lymphoid cryosections by applying E or EAC rosette-forming procedures and the membrane immunofluorescence technique. The receptor of B lymphocyte for C3 was revealed to be quite stable even in cryosections. The B lymphocytes were localized mainly in lymph follicles, above all in germinal centers, of lymph nodes and palatine tonsils. In addition, the lymph follicles were composed mostly of IgM and IgG-bearing lymphocytes on their cell membranes. The rosette-forming capacity of T lymphocytes with E in cryosections was rather unstable compared with that in suspended cells. The capacity to keep E on the T lymphocytes in cryosections was strengthened, however, by adding centrifugal force between the incubations at 37℃ and at 4℃. According to this newly developed technique, the E adhered diffusely to the cortex and medulla of the thymic section. The T lymphocytes were also demonstrated to localize in the paracortical area of the lymph nodes and the interfollicular region of the tonsils. Neither B lymphocytes, macrophages, monocytes nor activated T lymphocytes could bind EA (IgG) on lymphoid sections. Almost all of the lymphocytes invading the salivary glands of Sjogren's syndrome were revealed to be B lymphocytes, while T lymphocytes were localized periductally. Most of the infiltrating lymphocytes in Hashimoto's chronic thyroiditis were identified similarly as B lymphocytes. The T lymphocytes could be found mainly around the thyroid epithelium. In the spleen of patients with hypoplastic anemia, it was demonstrated that the white pulp consisted of B lymphocytes, and the red pulp is composed predominantly of monocytes having an Fc receptor. In addition, the white pulp was shown to be formed of lymphocytes bearing IgG on their surfaces. The E-RFC, i.e. T lymphocytes, were not detected in this spleen. The non-thymomatous thymus of patients with myasthenia gravis had lymph follicles which consisted of B lymphocytes like those of the peripheral lymphoid tissues, while the T lymphocytes were observed in the cortex, except for the lymph follicles produced.
