Freeze-etched Surface of Human Erythrocyte Membrane : Effect of the Anti-membrane Antibody

The anti-membrane antibody in serum of patients with systemic lupus erythematosus was found by electrosyneresis as reported previously. Further elucidation of the function of the antimembrane antibody in vitro and in vivo was performed using freeze-etched electron microscopy. The erythrocyte membranes were prepared from normal subjects and patients with systemic lupus erythematosus. The anti-membrane antibody was isolated from a patient with systemic lupus erythematosus and coupled to ferritin with p,p'-fluoro-m,m'-dinitrodiphenil sulfone as conjugation reagent. Ferritin-conjugated anti-membrane antibody was demonstrated on the surface of erythrocytes by freeze-etched electron microscopy. While the particles on the erythrocyte membrane of normal subjects are 85 A° (ranging from 40 to 140 A°) in diameter and 3,600/μ^2 in density, the particles on the membrane of patients with systemic lupus erythematosus increased their diameter to 130〜220 A°and decreased their density to 170〜450/μ^2. Furthermore, the particles on the erythrocyte membrane of normal subjects showed larger sizes and lower density, 150 A°(ranging from 30 to 200 A°) and 650/μ^2, respectively, after reacting with anti-membrane antibody coupled with ferritin. From these observations, it can be concluded that the anti-membrane antibody exists in sera of patients with systemic lupus erythematosus and reacts with the membrane-associated glycoprotein in vivo as well as in vitro.
