Analysis of Nucleoid-Proteins in Tobacco Chloroplasts

Highly purified chloroplast-nucleoids were isolated from cultured green Nicotiana tabacum cells. Electron microscopic observation indicated that these nucleoids had a compact, beaded structure. Disruption of the structure by protease or 2 M NaCl was consistent with the notion that the nucleoids were composed of chloroplast DNA and proteins held together by ionic interaction. The nucleoids showed run-on transcriptional activity in vitro. Neither nuclear DNA nor histone were detected in the nucleoid fraction. About 25 chloroplast-nucleoid proteins were characterized by 2D-PAGE, and DNA binding proteins were detected by 2D-PAGE and southwestern blotting. The subunit molecular mass of one DNA binding protein was 41 kDa and the pI was basic. The 41 kDa DNA binding protein was also detected in etioplast. The role of this 41 kDa DNA binding protein is discussed.
