Immuno-gold Localization of Nitrate Reductase in Spinach (Spinacia oleracea) Leaves

The intracellular location of nitrate reductase in spinach leaves was examined by applying an immunocytochemical method. Thin sections were first treated with immunopurified anti-nitrate reductase monospecific antibodies, followed by incubation with colloidal gold-labelled goat anti-rabbit immunoglobulin G as a marker. The nitrate reductase was specifically located in the chloroplast. When anti-nitrate reductase antibodies were omitted, or when pre-immune serum was used no label was observed.
