Three dimensional analysis of capillary in the islet of Langerhans of developing rat pancreas

Three-dimensional relationship of capillary network to endocrine cells was examined in the developing islet of Langerhans by a computeraided reconstruction from serial section light micrographs, and result was compared with those of mature pancreas. Rats from 0 to 7 days after birth were fixed by perfusion through the left ventricles with Zamboni's solution followed by preparation of serial sections with 3um thickness for paraffin and 1um for epoxy embedded samples. A part of samples was prepared for electron microscopy. Insulin cells were marked by immunohistochemistry with C-peptide antibody. The 3-D reconstruction was made with a use of TRI computer soft ware (RATOC Co). In the pancreas at 5 to 7-day-old rats, islets with varying size ranging 30-200um in diameter could be recognized. In the smaller and immature islet, capillary passed uniformly through the islet. In contrast, one thick capillary passed centrally in the maturated islet with surrounding capillary basket at the exterior of the islet, and there were several communications between central and outer ones. Endocrine cells have been shown to maintain intimate topological relation with the developing capillaries. The mode of capillary development was quite similar to that in regeneration after the 90% pancreatectomy. The evidence strongly indicated that the proliferation of endocrine cells cooperated with vascular neogenesis.
