Effect of pH on the Dimorphic Crystal Growth of Lysozyme

pH-dependence of the crystal growth of lysozyme chloride was observed at 15and 40'C. The size of tetragorral crystals grown at I5'C varied considerably withpH, while that of orthorhombic ones obtained al. 40'C showed hardly any pH-dependence in the range of pH 4-6.5. The results seem to indicate th'at (l) pH-insensitive hydrophobic interaction plays a greater role in the high-temperaturecrystallization, (2) the conformational changes at about 25'C found by NMRand enzyme kinetics to occur near the active site are not directly reflected in thecrystal growth and (3) the role of water must be taken into account as a primarycause of the dimorphism.
