Measurement of Bone Calcium and Phosphorus in Growing Rats By X-ray Fluorescence Analysis

The mineral contents in the growing rat bones are well known to change quantitatively depending on many factors. The object of this experiment was to study the effect of growth on changes in the calcium and phosphorus contents in the bone. In this study we used 4 week-old female rats fed on a normal diet continuously throughout the experiment. The mandibles and femurs were extracted at 4 weeks, 6 weeks and 8 weeks of age. The bones were fixed in a 10% formalin solution, washed with distilled water, and air-dried. Bone contents were determined at the following sites : The head of the mandible (mandible I), the region including the teeth anterior to a line perpendicular to the tangential line drawn at the inferior margin of mandible (mandible II) and the head of the femur (femur). The bone sample was pulverized and the resulting powder was shaped in a ring under pressure. The calcium and phosphorus contents in the samples were then analyzed quantitatively using an X-ray fluorescent analyzer (ZSX101e Rigaku Industry). The calcium and phosphorus contents in mandible I did not increase with growth. The contents in mandible II and femur tended to increase with growth. ZSX101e is widely used for quantitative analysis of food and mineral contents. The results of the present study showed similar tendencies to those analyzed by the traditional and widely used DXA method. Our findings showed that the analyzer is useful for quantitative determination of bone contents. Mandible I showed no increase as the rats aged, probably because growth of the mandible head begins later than age 8 weeks in these rats. We shall further examine the changes in contents in bone over time in a larger number of animals for a longer period of time.
