The effect of human lymphoblastoid interferon on fibronectin production was investigated in three breast cancer cell lines, two fibroblast cell lines, and each of fibroadenoma, mammary epithelial cells and milk epithelial cells. In all the cells, interferon inhibited cell proliferation, and caused an increase in cell volume. Fibronectin produced by the cells was divided into the two groups, cell-associated fibronectin and fibronectin released into medium, and was determined separately by radioimmunoassay. Production and retention of fibronectin were evaluated for cell protein content. The producing ability declined in fibroblasts, fibroadenoma and epithelial cells and was inhibited in one breast cancer cell line. On the other hand, changes in fibronectin production per cell revealed that fibronectin producing ability was inhibited in all the epithelial cells by interferon, although it was accelerated in fibroblasts. No changes were observed in cancer cells. As for the ratio of cell-associated fibronectin to total fibronectin, interferon decreased cell-associated fibronectin in fibroblasts and cancer cell lines, but increased it in epithelial cells.
