The clearance of exogenous recombinant human interleukin-2 (hIL-2) was characterized in a dog model. The titer of recombinant hIL-2 activity was measured using a radioimmunoassay. Intravenously injected hIL-2 disappeared from the blood circulation rapidly with a serum half life of 4.5±0.1 minutes (mean±SD). The reason for the short half life of i.v. injected hIL-2 was studied. To determine whether hIL-2 has been bound or consumed by peripheral blood lymphocyte (PBL) or serum, the IL-2 titer that has been cultured with PBL or serum in vitro was measured. No inhibitable effect of either lymphocyte and serum on the initial titer of cultured IL-2 could be detected. Interestingly, a surgical bilateral renal block on the dog greatly extended the serum half life of hIL-2 to 33.9±2.7 minutes. Administered hIL-2 was not excreted in a detectable form in the urine either the control or experimental group. These findings demonstrated that the kidney is the major organ to metabolize IL-2,possibly renal tubular catabolism.
