Production of abnormal prothrombin(PIVKA-II) by human hepatoma cells in culture.

Abnormal prothrombin PIVKA-II was detected and increased with time in cultures of human hepatoma cell lines huH-1, huH-2, PLC/PRF/5, HepG2 and Hep3B without added vitamin K. There was a marked decrease of PIVKA-II with added vitamin K. PIVKA-II was not detected in cell cultures AZ-521, MKN-7 (gastric cancer) and MIA PaCa-2 (pancreatic cancer). Thus, PIVKA-II is produced by hepatoma cells and its production is dependent on the amount of vitamin K available (in medium or in cell).<BR>All 3 enzymes involved in the production of prothrombin, vitamin K-epoxide-reductase, vitamin K-reductase (vitamin K cycle) and γ-glutamyl-carboxylase, functions in hepatoma cell huH-2. Thus it was suggested that production of PIVKA-II by huH-2 was due to increased formation of prothrombin precursors that resulted in an imbalance between the vitamin K cycle and γ-carboxylation-system with a relative vitamin K deficiency.
