Detection of hepatitis B virux X-gene protein and anti-X in type B chronic liver disease.

The genome of the hepatitis B virus contains a sequence (ORF X), the role of which is unclear. The aimost compiete region of HBV X region was expressed in Escherichia coli, and the resulting protein was about 17KD of MW. Rabbit polyclonal antisera against X protein was prepared.<BR>Sera of HBs-Ag positive 55 patients were analyzed by Western blot assay. Anti-HBX was present in 28% (7/25) sera with chronic hepatitis, in 52% (11/21) with liver cirrhosis and 56% (5/9) with hepatoma. The expression of HBV X product in liver tissue was investigated with indirect immunofluoresence method. X protein was observed in 69% (9/13) patients with chronic hepatitis, and in 50% (5/10) with liver cirrhosis. X protein was not expressed when one's serum was positive for anti-X.<BR>HBX protein in liver and anti-X in serum couid be detected in HBV marker positive patients. The expression of anti-HBX seemed to increase with the progress of liver disease. Presence of anti-HBX might suppress the expression of HBX protein in liver.
