Effects of Pd-Ia, a Component of Chinese Traditional Herb Bai-Hua Qian-Hu, on Ca^<2+>-Current and Action Potential Duration in Single Ventricular Cells of Guinea Pig Hearts

A Chinese herb, "Qian-Hu," is traditionally used for the treatment of common cold, chest pain and hypochondria. In this study, by the patch clamp techniques (whole cell recording) and conventional glass microelectrode methods, we examined the effects of Pd-Ia, a coumarin component isolated from Bai-Hua Qian-Hu (Peucedanum praeruptorum DUNN.; Umbelliferae), on the action potential duration (APD) and Ca^<2+>-currents in single ventricular cells of guinea pig hearts. Ten minute-superfusion with normal Tyrode solution containing 1 μmol/l Pd-Ia reduced APD_<20> and APD_<50> by 16.8 % and 8.3 %, respectively. Pd-Ia also decreased the Ca^<2+>-current in the single myocytes in a dose-dependent manner.
