On the Evaluation of the Preparation of Chinese Medicinal Prescriptions (VII) : HPLC Analysis of the Components in "Inchinko-to"(茵〓蒿湯) and "Inchin-gorei-san"(茵〓五苓散)

This paper describes quantitative and qualitative analyses of some compounds in the Chinese traditional medicinal prescriptions, "Inchinko-to" (茵〓蒿湯) consisting of Inchinko, Gardenia fruit and Rhubarb, and "Inchin-gorei-san" (茵〓五苓散) consisting of Inchinko, Alisma rhizome, Chuling, Iloelen, Atractylodes rhizome and Cinnamon bark, by three dimensional (3D) HPLC. First, we found that 6,7-dimethylesculetin, capillarisin, geniposide and sennoside A in an "Inchinkoto" model preparation were clearly separated from each other and identified by 3D-HPLC. Then, this 3D-HPLC method was applied to the analysis of 6,7-dimethylesculetin, capillarisin and cmnamic acid in decoctions of "Inchin-gorei-san." The characteristic compounds of all crude drugs used in "Inchinko-to" were identified. In case of "Inchin-gorei-san," the characteristic compounds of Inchinko, Atractylodes rhizome and Cinnamon bark were identified by the 3D-HPLC but none of the specific compounds of Alisma rhizome, Chuling and Hoelen was detected. The result of the present quantitative analysis showed that the availability (移行率) of 6,7-dimethylesculetin was 46.7 %, 32.9 % and 74.3 °/ for Inchinko water ext., "Inchinko-to" and "Inchin-gorei-san," respectively, and that of capillarisin, 10.0%, 16.3% and 43.5% for Inchinko water ext., "Inchinko-to" and "Inchin-gorei-san," respectively. This method is considered to be useful for the chemical evaluation of "Inchinko-to" and "Tnchin-gorei-san."
