Effect of Oren-gedoku-to on Change of the Hexobarbital-Induced Sleep Time in Diazepam-Treated Mice

The effects of Oren-gedoku-to on the hypnotic duration induced by hexobarbital, and on the change of the duration in diazepam-treated mice were examined, A single dose of Oren-gedoku-to (1.0 g/kg, p.o.) or diazepam (1.0 mg/kg, p.o.) significantly prolonged the hexobarbital-induced hypnosis time. When diazepam (1.0 mg/kg, p.o.) and Oren-gedoku-to (1.Og/kg, p.o.) were given simultaneously, the hypnosis time was longer than that produced by administration of Oren-gedoku-to or diazepam alone. Coptis rhizome and Gardenia fruit (each 600 mg/kg, p.o.), component crude drugs in Oren-gedoku-to showed the effect similar to that of Oren-gedoku-to itself on hexobarbital-induced sleep time. These effects were antagonized by flumazenil (15.0 mg/kg, p.o.), a benzodiazepine antagonist.
