<Poster>Involvement of P2X2 and P2X3 receptors in dorsal root ganglion neurons of streptozotocin-induced diabetic neuropathy

Painful diabetic neuropathy causes allodynia and does not respond to commonly used analgesics such asnon-steroidal anti-infl ammatory drugs or opioids at doses below those producing disruptive side eff ects. In the presentstudy, we examined the effect of P2X receptor antagonists, which are known to modulate the pain pathway, onmechanical allodynia in streptozotocin( STZ)-induced diabetic mice. The paw withdrawal frequency measured by vonFrey fi laments, began to signifi cantly increase 5 days after STZ injection and was maintained for more than 14 days.Intrathecal administration of P2X receptor antagonists (PPADS and TNP-ATP) inhibited the mechanical allodyniain diabetic mice. Next, the levels of mRNA for the P2X receptors in DRG in diabetic mice were measured usingquantitative real-time PCR. The levels of P2X2 and P2X3 receptors mRNA were signifi cantly increased in diabeticmice at 14 days after the intravenous injection of STZ. Furthermore, we investigate the localization of glial cells andneuron in spinal cord of diabetic mice. However, microglia, astrocyte and neuron were not changed in spinal cord.These results suggest that the upregulation of P2X2, P2X3 and/or P2X2/3 receptor in DRG neurons is associated withmechanical allodynia in STZ-induced diabetic mice.
