Stimulatory Effect of Medroxyprogesterone acetate on p27 Protein in Endometrial Cancer Cells

p27 is regarded as a cyclin dependent kinase inhibitor of the G1-to-S cell cycle progression. This study was designed to investigate whether progestin, medroxyprogesterone acetate (MPA), accumulated p27 protein in human endometrial cancer cell-lines, HEC-265 and HEC-50B. HEC-265 was positive for progesterone receptor (PR) and HEC-50B was negative for PR by immunocytochemical staining. Quantitative analysis of p27 protein was performed by immunoblotting. Maximum effect of MPA on p27 accumulation in HEC-265 was observed at 1×10^<-6> M. MPA induced p27 protein level 1.56 times, 24 hours after addition of MPA, whereas no effect of MPA was realized in HEC-50B. MPA, 1×10^<-6> M and 1×10^<-5> M, suppressed HEC-265 cell growth. The reduction rate of MPA 1×10^<-6> M was 31% on day 6. These data suggest that p27 induced by MPA may be involved in the progesterone-induced growth suppression of human endometrial adenocarcmoma.
