An Experimental Renal Disease Induced by the Genetic Immunization of Alpha-2 Macroglobulin Receptor-Associated Protein cDNA

Experimental models of renal disease have been induced by the administration of putative proteins or pathogenic antibody for a long time. By using the recent advances of molecular biology, the establishment of the molecular-based experimental model of renal disease could be possible. To establish the molecular-based experimental model of renal disease. The expression vector pKA1 containing the monkey DNA of alpha-2 macroglobulin receptor-associated protein (α2MRAP), a Heymann nephritis-related antigen, administered to normal Lewis rats once a week until 29 weeks. Urinary protein excretion measured by using the biulet method, antibody production against α2MRAP in the sera by using the western blot analysis and the renal histological analysis were examined. A significant but minimal level of proteinuria appeared from 19 weeks and thereafter. Antibody against α2MRAP was produced in the sera after 6 weeks and maintained. Renal histology showed the shedding of α2MRAP from the glomerular podocytes at 29 weeks. The α2MRAP DNA-induced experimental model of renal disease has been established in the normal Lewis rat.
