Establishment and Characterization of a Novel Human Chronic Myelogenous Leukemic Cell Line, MT-12

A myeloblastic cell line, designated MT-12, was established from the peripheral blood of a patient with blastic crisis of Philadelphia (Ph^1) chromosome-positive chronic myelogenous leukemia. MT-12 cells grew in single cell suspension with a doubling time of about 30 hours. Chromosome analysis revealed 49XY, t (9; 22) (34q; 22q), +8, +8, +22q-. Undifferentiated MT-12 has the granulocytic cell surface marker. After the treatment of cell differentiation reagents, MT-12 cells gain both megakaryocytic and erythroblastic properties on the basis of cell staining and surface analysis, showing that MT-12 is a tripotent, megakaryocytic, erythroid, and granulocytic cell line. In contrast to platelets or Meg-01 (megakaryocytic cell line), MT-12 synthesized little amount of prostaglandins including thromboxane A_2 (TXA_2) by addition of thrombin or ionomycin even in megakaryocytic-differentiated state. MT-12 has a single class of TXA_2 receptor like platelets. TXA_2 agonists such as U46619 or STA_2 increased intracellular concentrations of Ca^<2+>([Ca^<2+>]i) more than other prostaglandins, its analogues or receptor-mediated cell stimulants. Thus, MT-12 is a useful probe to investigate the regulation of both megakaryocytic and erythroid differentiation, and the role of TXA_2 receptor or prostaglandin-related enzymes in cellular function.
