REGULATION OF ELASTIC FIBER COMPONENTS AND EFFECT OF TROPOELASTIN ON IN VITRO VASCULAR CALCIFICATION

Vascular calcification is a common feature of advanced atherosclerosis and results in reduced elasticity of elastic arteries. However, it remains unclear that the relationship between vascular calcification and elastic fiber in molecular and cellular levels. We investigated expression of major elastic fiber components, such as tropoelastin (TE), fibrillin-1 (FBN1), and lysyl oxidase (LO), on calcification model using β-glycerophosphate (β-GP) in bovine aortic smooth muscle cells (BASMCs). As determined by Western blot analysis, protein secretion of TE and FBN1 was time-dependently repressed by exposure to 10 mM of β-GP. Also, mRNA expression of elastic fiber components (TE, FBN1, and LO), as assessed by RT-PCR, was down-regulated on β-GP-induced calcification. In experiments using calcification inhibitors, we used an ALP inhibitor, levamisole, and a bisphosphonate, DMDP, as a calcification inhibitor. Both of levamisole and DMDP inhibited the β-GP-dependent down-regulation of TE, FBN1, and LO mRNAs, as well as the β-GP-induced calcium deposition. Furthermore, we investigated whether TE can regulate the extent of vascular calcification in this model. When recombinant TE was added to BASMCs together with β-GP, recombinant TE dose-dependently attenuated the calcium deposition. In conclusion, it is suggested that elastic fiber components, TE, FBN1, and LO are down-regulated on mRNA levels on in vitro calcification models and that the down-regulation of TE may directly relate with calcium deposition.
