An Oxygen-Evolving Photosystem II Complex Associated with the Core Substructure of the Phycobilisome from Synechococcus elongatus

Oxygen-evolving photosystem II (PS II) particles isolated from the thermophilic cyanobacterium Synechococcus elongatus consisted of about twenty polypeptides. Six polypeptides were identified by reaction with specific antisera as constituent subunit polypeptides of oxygen-evolving PS II reaction center complexes. The most abundant polypeptides were the a and p subunits of allophycocyanin. Comparison with the polypeptide profile of isolated phycobilisomes, as well as immunoblotting with an antiserum against the large linker polypeptide, showed that the large linker polypeptide or some proteolytic fragments of it were present in the preparation. Thus, each PS II particle is, in essence, an oxygen-evolving PS II complex that is associated with the core substructure of the phycobilisome. Cross-linking experiments indicated that fragments of the large linker polypeptides are closely associated with one another and that the Chl-carrying 47-kDa polypeptide is located in close proximity to the D2 protein and the extrinsic 33-kDa protein.
