Studies on cytochromes in photosynthetic electron transport system I. Photoreduction and photo-oxidation of cytochrome b_<559> by photosystem II in spinach chloroplasts

Light-induced redox-reactions of cytochrome b_<559> in spinach chloroplasts were investigated. Illumination of chloroplasts induced photoreduction of cytochrome b_<559>. Red light (650 nm) was more effective than far-red light (725 nm), indicating that the photoreduction is a photosystem II-mediated reaction. On addition of DCMU, the photoreduction was eliminated and a photooxidation of cytochrome b_<559> was observed. The rate of this photooxidation was faster with photosystem II light than with photosystem I light. On addition of Mn^<++>, the photooxidation was partly suppressed; far-red light became as effective as red light in inducing photooxidation of cytochrome b_<559> in the presence of DCMU and Mn^<++>. Ascorbate completely suppressed photooxidation of cytochrome b_<559>. In the presence of ascorbate, however, photooxidation was observed in the presence of inhibitors or after inhibitory treatments of chloroplasts which affected the oxidizing side of system II. These inhibitors and inhibitory treatments, but not DCMU, decreased the redox-potential of cytochrome b_<559>. Reactivation of Hill reaction in Tris-washed chloroplasts by indophenol-ascorbate treatment was not accompanied by an abolishment of photooxidation of cytochrome b_<559>. A possible mechanism is proposed to account for these reactions of cytochrome b_<559> m the photosynthetic electron transport in chloroplasts.
