Effects of auxin on the structure of hemicelluloses of Avene coleoptiles

Avena coleoptile hemicelluloses were fractionated into water-soluble hemicelluloses I and IIB and water-insoluble hemicellulose IIA. These hemicelluloses were then subjected to glycosidic linkage analysis by methylation technique, which revealed that hemicellulose I was predominantly composed of arabinoxylans and β-(1→4) glucans and hemicellulose IIB was composed of arabinoxylans, p-(1→4) : (1→3)-mixed linked glucans, β-(1→4)-glucans and xyloglucans. Hemicellulose IIA was mainly composed of xyloglucans and probably β-(1→4)-glucans. Methylation analysis of hemicelluloses extracted from Avena coleoptile segments treated with auxin in the presence of mannitol (0.15 M) indicated that auxin apparently had no effect on the structure of arabinoxylan and caused a specific decrease in the amount of β-(1→4) : (1→3)-mixed linked glucan.
