Auxin-induced extension, cell wall loosening and changes in the wall polysaccharide content of barley coleoptile segments

Contents of the cell wall and sugar pool and the response to exogenously applied auxin (cell extension and cell wall loosening) were investigated with barley coleoptile segments excised from 4-, 5- and 6-day-old seedlings. The first two groups exhibited a high capacity to grow in terms of the intact growth rate and were responsive to auxin, while those excised from 6-day-old seedlings had a low growth capacity. The cell wall of 4- and 5-day-old coleoptile segments contained almost the same amount of noncellulosic wall components per unit length while the 6-day-old ones had a lesser amount. The sugar pool and α-cellulose content per unit length decreased as the coleoptile aged. Auxin-stimulated extension was most marked in the 4-day-old coleoptile segments. Auxin caused quantitative changes in the cell wall components of 4-day-old coleoptiles and, to a lesser extent, of 5-day-old coleoptiles, i,e., an increase in the contents of xylose and arabinose, both of which are constituents of noncellulosic polysaccharides of the cell wall, and of α-cellulose and a decrease in the noncellulosic glucose content. Auxin caused very little change in the noncellulosic sugar content and α-cellulose content of the cell wall from 6-day-old coleoptile segments. The auxin-induced change in mechanical properties of the cell wall was significant in 4- and 5-day-old coleoptiles but very small in 6-day-old ones. The results suggested that the content of noncellulosic wall components is closely related to the intact growth and auxin responsiveness of barley coleoptiles.
