The effect of IAA and FC on the extension of isolated epidermis of light-grown Alaska pea epicotyls was studied under different conditions with an extension apparatus. The following results were obtained. (1) The epidermis extended in response to low pH buffer solution of 1-10 mM, maximum extension being achieved at pH below 5.5. (2) IAA, 5 mg/liter, caused, although not consistently, an extension of epidermal strips in 1 mM buffer, but not at 10 mM. (3) Consistent extension of the isolated epidermis due to IAA was obtained by addition of GTP, ATP. ITP or UTP (sodium salts), but not nucleosides, nitrogen bases or sugars. (4) A fungal toxin, FC, at 10^<-5> M induced extension of the epidermis without addition of the nucleoside triphosphates. (5) IAA and FC caused H+ extrusion in peeled epicotyl segments both in the presence and absence of GTP. IAA caused appreciable H^+ extrusion in the isolated epidermis only in the presence of GTP, whereas H^+ extrusion by the epidermis was induced by FC even in the absence of GTP. From these results, we concluded that IAA induces extension of the isolated epidermis under the above conditions through the mediation of H^+ ions.
