Isolation and Characterization of a Fast-Growing, Thermophilic, Hydrogenotrophic Methanogen

A very fast-growing, thermophilic, autotrophic methanogen (strain KN-15) was isolated from mud around a hot springs. The bacterium KN-15 which was obligately anaerobic, Gram-positive, and non-motile rod, could utilize only H_2-CO_2 gas as carbon and energy sources. The optimum growth temperature and pH values were 63° to 66℃ and 7.4 to 7.8,respectively. The bacterium KN-15 was the fastest-growing thermophilic rodshaped methanogen among the isolates reported under low NaCl concentration (0 to 5 g・l^<-1>), and had a specific growth rate of 0.62h^<-1>. The G+C content of DNA in teh KN-15 was 47.6±0.5 mol%. KN-15 was determined to belong to the genus Methanobacterium.
