Substrate-Microbial Behavior and Kinetics in the Anaerobic Digestion of Night Soil : Kinetic Studies on the Anaerobic Digestion of Night Soil (II)

The time course of substrate removal and microbial growth in the batch and semicontinuous processes of the anaerobic digestion of synthetic night soil at 35℃ and at different sludge retention times was investigated to elucidate the reaction mechanism between substrate and microbe using the Michaelis-Menten equation and its modified form for an enzyme reaction.The plots of the specific substrate removal rate, (1/S)(dl_r/dt), against the modified substrate equilibrium concentration, l_e/S, show that the reaction velocity curves are better fitted to the modified Michaelis-Menten equation which is expressed as : 1/S((dl_r)/(dt))=(k・(l_e)/S)/([K_m]+(l_e)/S) where (1/S)(dl_r/dt)=specific substrate removal rate (g/g/day); k=maximum specific substrate removal rate (g/g/day); [K_m]=the modified Michaelis-Menten constant; l_e=substrate equilibrium concentration (mg/l); and S=microbial concentration (mg/l)/Biokinetic constants which are required in the formulation for the design and control of batch and semicontinuous processes of anaerobic digestion of night soil were also determined from the developed kinetic models. Kinetic models that interrelate the specific microbial growth rate, (1/S)(dS/dt), the specific substrate removal rate, (1/S)(d;_r/dt), and the specific gas productino rate, (1/S)(dG/dt), as well as the hydraulic retention time against the specific growth rate were also established, viz. 1/S((dG)/(dt))=a・1/S((dl_r)/(dt))+b' 1/S((dG)/(dt))=a'・1/S((dS)/(dt))+b" 1/S((dS)/(dt))=c・υ/V where a, b', a', b" and c are experimental kinetic constants, υ is the daily night soil input or the daily wastage of the digester mixed lipuor and V is the digester volume.
