Stereoselective Conversion of Isobutyric Acid to β-Hydroxyisobutyric Acid by Microoganisms : Studies on β-Hydroxycarboxylic Acids (I)

The stereoselective β-hydroxylation of isobutyric acid or methacrylic acid by microorganisms was studied. A high activity for the β-hydroxylation was observed in Candida rugosa, Hanseniaspora valbyensis, Hansenula anomala, Trichosporon aculeatum, Tr. fennicum, Endomyces reessii, Geotrichum loubieri, Micrococcus flavus, M. luteus and M.lysodeikticus. And while some microorganisms, for example, C. rugosa IFO 0750,IFO 0591,C. parapsilosis IFO 0708,Pichika membranaefaciens IAM 4904 and Torulopsis candida, produced the D-form of β-hydroxyisobutyric acid, most accumulated exclusively L-β-hydroxyisobutyric acid. The stereoisomeric configulations of β-hydroxyisobutyric acid were confirmed by specific optical rotation, NMR and IR spectra. The specific optical rotation of D- and L-β-hydroxyisobutyric acid obtained by use of Candida rugosa IFO 0750 and IFO 1542 was [α]^<35>_D -17.6° and 17.8° (C=1,MeOH), respectively, while that of their methyl esters was [α]^<25>_D -25.2°and +25.0° (C=4,MeOH).
