Histological study of a paclitaxel-induced peripheral neuropathy model treated with goshajinkigan

Paclitaxel is an effective anticancer drug for treatment of gynecologic cancer, but it is associated with peripheral neuropathy. We prepared a rat model of continuous administration of paclitaxel directly to the sciatic nerve (paclitaxel model), and confirmed that orally administered goshajinkigan inhibited paclitaxel-induced neuropathy. In this study, we examined the sciatic nerve of the paclitaxel model treated with goshajinkigan, and histologically investigated why goshajinkigan decreased neuropathic pain. Male Sprague-Dawley rats were divided into the paclitaxel group and the paclitaxel plus goshajinkigan group. One, two, three and four weeks after surgery, sciatic nerve sections were prepared and observed under a light microscope. In addition, the number of myelinated nerve fibers per unit area was calculated, and the ratios of small-diameter to largediameter nerve fibers were compared. As a result, there is no significant histological difference between the two groups, and that goshajinkigan did not promote regeneration of nerve structures.
