The relative deoxyribonucleic acid content of cells in culture was determined by flow microfluorometry (F.M.F.) after staining with propidium iodide. Effects of daunorubicin (DNR), cytosine arabinoside (Ara-C), 6-mercaptopurine (6MP) and prednisolone (Pred) used broadly as antileukemic drugs on cell cycle progression were investigated by F.M.F.. Exposure of cultured RPMI 3236 cells to 0.5μg/ml of DNR for 1 hr led to an accumulation of cells at late S and G_2 phases until 48 hrs of incubation. Continuous contact of cells with the drug caused no difference in DNA histogram comparing with control group. The result showed that DNR might have an action to inhibit the progression of cell cycle. In experiments of a short exposure of cells to antimetabolites (Ara-C and 6MP), a transient increase in cells at S phase was observed but after 48 hrs they returned to the original cell cycle. In the continuously treated experiment, cell population accumulated at whole S phase and gradually reached late S phase without cell growth, suggesting that these drugs had a time-dependent action. Continuous contact with Pred resulted in almost no change in DNA histogram, but a slight prolongation of S phase was observed. F.M.F. is very suitable for the study on effects of drugs on cell cycle progression and, because of its rapid measurement, seems to be one of methods applicable in the treatment of acute leukemia.
