Increased expression of the mts1 gene in urogenital cancers

Objective : Fluorescence differential display (FDD) is a useful method for detecting differences in gene expression patterns between disease and healthy control subjects. When FDD was first applied to the detection of abnormally expressed genes in prostate cancer, we found the genes mts1 (also known as CAPL or S100A4) and lactoferrin previously. In this paper we evaluated the amount of expression of these genes in prostate cancer (CAP), renal cell carcinoma (RCC), and bladder tumor (BT), using Real-time PCR. Materials and Methods : Semiquantitative reverse transcription polymerase chain reaction (PCR) was performed in surgically excised specimens of CAP, RCC, and BT and compared with specimens from control subjects to evaluate the possible cancer-specific expression of these genes. Real-time PCR for these genes was then conducted in RCCs and BTs and compared with specimens from control subjects in larger samples to evaluate grade and stage dependency. Results : The expression of nts1 was higher in all cancer samples examined compared with the control samples. The expression of lactoferrin was not different in any cancer sample compared with the control samples. A possible association of increasing mts1 expression with the cancer stage and grade was recognized in BTs. Conclusion : FDD might be useful for researching cancer-specific genes. The expression of mts1 was higher in examined urogenital cancers than in control specimens, which indicates the potentially active role of the mts1 gene in the progression of these malignancies.
