Absence of a Mutation of the Ras Genes in Human Prostate Cancer

Various human tumors have been studied for ras gene mutations to date, but data reported for prostate cancer do not always accord well. Prostate cancer cell lines and three American studies on prostate cancer demonstrate low incidence of the ras mutation. But Japanese clinical, latent and autopsy specimens show an approximately 24% incidence of H- and K-ras mutations. We examined ras gene mutation by polymerase chain reaction single-strand conformation polymorphism and direct sequencing analysis in 34 primary clinical prostate cancers, 1 metastatic lymph node, 2 prostate cancer cell lines (PC3 and DU145) and 2 benign prostatic hyperplasia samples. No mutation could be found at codons 12, 13, or 61 of H-, K- and nor were there any N-ras genes in any samples studied. The results suggested that the ras genes mutation is not associated with carcinogenesis in clinical prostate cancer and the incidence of the ras gene mutation in prostate cancer may not differ between American and Japanese males.
