An Application of the Fluorescence Differential Display Method to Clone Genes Related to Prostate Cancer

Multiple genes are involved in the development and the progression of malignant tumors, and extensive studies for a variety of tumors are revealing the genes involved in specific tumor formation. In the present study, we used the fluorescence differential display (FDD) method to investigate the genes involved in the development of prostate cancer. Total messenger RNA fraction was extracted from the prostate cancer tissue of a patient and the FDD method was employed. A total of 96 bands, longer than 300bp, were isolated as the bands expressed specifically in the cancer tissues but not in the comparative total RNA derived from the testicular tissue. Further processing of these bands gave 18 cDNA subclones, from which 3 cDNA subclones were worked up to obtain complete nucleotide sequences. A BLAST search indicated that these genes were identical to lactoferrin, CAPL/mstl and 3-phosphoglycerate-dehydrogense. A study of the CAPL/mtsl gene showed significantly higher expression in prostate cancer compared to benign prostate hypertrophy suggesting that the CAPL/mtsl gene is involved in the proliferation and the progression of prostate cancer. However, the expression of lactoferrin was the same in both prostate cancer and benign prostate hypertrophy.
