Fukuzawa et al. previously hypothesized that there is a definite minimum amount of disintegrator required for the disintegration of a directly compressed tablet which contains a large quantity of a drug insoluble or slightly soluble in water. That is, there is a critical disintegrator amount, above which a continuous structure of disintegrator particles is formed. The authors conjectured that some marked changes take place in the internal structure of the tablet above this critical disintegrator amount, and make it possible for the tablet to disintegrate. We therefore studied the changes of the internal structure and mechanical strength of tablets in order to elucidate the mechanisms involved. It was found that a critical disintegrator amount does indeed exist, and a continuous structure of disintegrator particles is formed, confirming the hypotheses of Fukuzawa et al. Enlargement of the pore diameter of tablets caused by the continuous structure of disintegrator particles may be a major contributing factor.
