Euglena gracilis z cultured in the vitamin B_<12>-deficient medium suffered more extensive digestion and gave higher yield of mitochondria than the cells grown in the vitamin B_<12>-sufficient medium. The Euglena mitochondria showed the highest respiratory control ratio and P/O ratio in the assaying medium containing 0.25M sucrose at pH 7.4. Electron transfer inhibitors inhibited the respiratory rate in state 3 and uncouplers stimulated the state 4 respiration. Oligomycin, which is an energy transfer inhibitor, inhibited the state 3 respiration of Euglena mitochondria in the identical degree as the mammalian mitochondria.
