Studies on Reactivity of T Lymphocytes Derived from Lewis Rats Undergoing Experimental Autoimmune Myocarditis (EAM)

Lymphocytes recovered from lymph nodes and hearts of Lewis rats immunized with porcine cardiac myosin (CM) were examined for their reactivity to the antigen. The cells recovered from lymph nodes responded to porcine CM by producing interferon-gamma (IFN-γ). Helper T (Th) 2 type specific T cells were not detectable. After culturing in vitro, the cells showed the response of the same specificity but required the addition of fresh antigen presenting cells. Furthermore, the cells crossreacted with the self antigen, i. e., rat CM. The induction of auto-crossreactivity to rat CM might explain the induction of autoimmunity. On the contrary, however, CD4^+ T cells recovered from the diseased hearts and expanded in vitro responded to porcine CM without showing detectable crossreactivity to rat CM. Although the pathogenic contributions of the observed T cell reactivities are not clear yet, this report is the first to determine the specific reactivities of the T cells obtained from experimental autoimmune myocarditis (EAM) rats particularly that of the infiltrating T cells.
