Alkaline Phosphatase activity revealed by undecalcified frozen sections of the periodontal ligament of rat molars during tooth movement

The aim of this study was to determine the distribution of ALP activity both in the normal PDL and in the PDL during tooth movement in rats. ALP activity was determined through examination of undecalcified frozen frontal sections of the whole PDL of the upper first molars. In the normal PDL, ALP activity was distributed throughout the periodontal ligament and in the connective tissue underlying the inner epithelium of the gingiva. Intense activity was observed along the periosteal bone surface and high activity was also observed on the periodontal bone surface. During tooth movement, ALP activity was increased on the periodontal bone surface in the tension zone. No significant change was observed in ALP activity at the cementum surface. In the pressure zone, no activity was observed in the degenerating tissues or on the adjoining bone surface. Intense activity was, however, observed on the remaining periodontal bone surface. The ALP activity in the remainder of the PDL, other than the degenerating tissues, was not greatly changed. The results demonstrate that ALP activity is affected by orthodontic stimuli. Elevation of ALP activity in the biochemical data is most likely due to the increase in PDL cellularity, or more specifically an increased in the number of osteoblasts with enhanced osteogenic activity.
