The periodontal ligament (PDL) of hypofunctional tooth shows atrophic changes. This study examines the effects of orthodontic force on the atrophically changed PDL. Atrophic changes of the PDL of upper left first molars were induced by removing the three antagonistic crowns in rats (Wistar, 13 weeks, male). After 3 weeks, the first molars with atrophied PDL were moved palatally for 3, 7, 14, 21 and 28 days, and then they were processed for histological examination. The atrophied PDL recovered from the start of the tooth movement to the 14th day. On the 3rd day, the periodontal width broadened, the osteoblasts increased, and the periodontal fibers projected out from the root surface. On the 7th day, the bone surface became rough due to newly formed bony spicules. The fibrous connection formed but each fiber was very thin. On the 14th day, the fiber network architecture was reorganized firmly, though each fiber less interlaced and was thinner than that of the normal group. From the 21st day, with decrease in the orthodontic force, the PDL showed a tendency of return to atrophic condition. Even atrophied PDL responds rapidly to orthodontic force by changing its structure. And, the extent and pattern of its response depend on the magnitude and direction of applied force. From these results, we conclude that the PDL possesses high adaptability to changes in the environment and thus always develops a structure suitable for its environment.
