On the Adsorption Mode of Tetracycline in Hard Tissue

This study was performed to examine proteins which have an affinity to tetracycline and the role of calcium ions in the reaction. Spectrophotometry was employed for observations of the optical density of tetracycline in calcium ion solutions of various concentrations, then the adsorbtion between tetracycline and proteins was examined using porcine enamel proteins which were electrophoretically transferred from polyacrylamid gels to nitrocellulose sheets. In spectrophotometry, the absorbance value increased in accordance with increases in the ratio of calcium ions against tetracycline until the level of about 1:500 where the value stayed on a plateau. In the adsorption experimet clear fiuorescence bands were recognized in the enamel protein bands corresponding to molecular weights, enamel proteins of 32 kDa, 13 kDa and smaller molecules. These results suggest that the intensity of tctracycline fluorescence depends on the concentration of the calcium ions and that tetracycline forms a complex with some proteins.
