Metabolism of [^<14>C]GA_<20> in a Cell-Free System from Developing Seeds of Phaseolus vulgaris L.

The metabolism of [^<14>C]GA_<20> during seed maturation of Phaseolus vulgaris was studied using cell-free systems from embryos ranging in age from 10 DAF (day after flowering) to 24 DAF. Enzyme preparations from very immature seeds actively converted GA_<20> to GA_1, GA_5 and GA_6. The ratio of incubation products suggested the biosynthetic pathway of GA_<20>-GA_5-GA_6. Fluctuation in the levels of endogenous C_<19>-GAs, namely GA_1, GA_4, GA_5, GA_6, GA_8, GA_9 and GA_<20> was analyzed by GC-SIM using internal standards to compare enzyme activity with the levels of endogenous GAs. Although GA_1, GA_4 and GA_6 Showed maximum levels on 20 DAF, enzyme activity decreased during seed maturation and showed weak activity on 20 DAF.
