Effects of Temperature on Growth and Protein and RNA Syntheses of Excised Vigna mungo Embryonic Axis

Embryonic axes excised from dry Vigna mungo seeds cultivated on wet filter paper grew well at 27℃, but did poorly below 15℃. The increase in fresh weight at 27℃ was completely inhibited by both cycloheximide and α-amanitin. Excised axes could grow well at 15℃ only if cultivated at 27℃ for the first 12h. Also, excised axes cultivated at 15℃ for several days grew well when transferred to 27℃. These results suggest that the ability of the axes to grow is retained during cultivation at 15℃, even when no growth occurs, and that only the initial stage of cultivation requires high temperatures for growth. The incorporation of ^3H-leucine into protein in the excised axes during the first 6 h of cultivation at 15℃ was much slower than that at 27℃. The incorporation rate at 15℃ during the second 6 h was, however, considerably high. In contrast, the incorporation of ^3H-uridine into the total RNA fraction at 15℃ was much slower during the first and the second 6 h as compared to that at 27℃, suggesting a difference in the response to temperature between protein and RNA syntheses in the axes.
