Effect of growth temperature on reserve mobilization and hydrolytic enzyme activities in Vigna mungo cotyledons

Seeds of Vigna mungo were allowed to germinate at 27, 18 and 15℃, and time-course changes of hydrolytic enzyme activities and the mobilization rate of reserve components in cotyledons were studied. The seeds germinated at 27 and 18℃ grew normally, whereas the growth at 15℃ was markedly retarded. In cotyledons of seedlings grown at 27 and 18℃, amylolytic and proteolytic enzyme activities increased at early stages of growth and the rates of starch and protein mobilization changed corresponding to the hydrolytic enzyme activities. At 15℃ the enzyme activities increased gradually during the experimental period of 16 days, but the reserves in cotyledons remained almost unchanged until the end of the experimental period. Changes of zymogram patterns of amylolytic and proteolytic activities in cotyledons of seedlings grown at 27, 18 and 15℃ were examined using polyacrylamide gel electrophoresis. The intensities of a main band of α-amylase and at least two bands of protease (gelatin-hydrolyzing activity) increased concurrently with in vitro activities of amylolytic and proteolytic enzymes. At least three bands of starch phosphorylase were present in cotyledons at early stages of germination and their intensities decreased during the growth of seedlings at 27, 18 and 15℃.
