Multiple Forms of Acid Phosphatase in Cotyledons of Vigna mungo Seedlings: Immunological Detection and Quantitation

Using a combination of column chromatography and gel electrophoresis, we have found that acid phosphatase in cotyledons of Vigna mungo seedlings is composed of at least six forms (Ia_1, Ia_2, Ib_1, Ib_2, IIa and IIb). We purified one of the major forms, Ia_1, as a polypeptide of 53 kDa. Using an antiserum raised against the enzyme Ia_1, we examined the immunological relationships between the multiple forms from cotyledons and the distribution of the enzyme in organs of maturing and germinating seeds.
