Stimulation of in Vitro RNA Synthesis by DNA-Loosely Bound Proteins Treated with Gibberellin A_3

Gibberellin A_3 (GA_3) treatment enhances the in vitro template activity of chromatin in peas (Pisum sativum, var. Alaska) (Tomi et al. 1983a). Effects of GA_3 on the chromatin template were surveyed for the chromatin components, DNA-tightly bound proteins (TBPs) and loosely bound proteins (LBPS). An addition of LBPS from GA_3-treated chromatin enhanced the template activity of LBP-depleted chromatin. In addition, both components of the LBPs, high mobility group proteins (HMGS) and low mobility group proteins (LMGS) were required for this enhancement. The LBPS converted LBP-depleted chromatin into a form susceptible to DNase I. LBPS probably are at least partly responsible for GA_3-enhanced template activity due to their relaxing the inter-action of core histones and DNA.
